BCR-ABL MutaPrime T315I RQ Kit (24 tests) is developed to detect the T315I mutation of the tyrosine kinase domain of the BCR-ABL1 chimeric gene in the clinical material of patients by means of polymerase chain reaction (PCR) with real-time hybridization-fluorescence detection.

The test system is compatible with amplifiers: "iQ5" (Bio-Rad); "CFX96" (Bio-Rad); "ABIPrism" (ThermoFisher Scientific); "Lightcycler" (Roche); "Rotor-Gene" 3000/6000 (Corbett Research/Qiagen).

The advent of tyrosine kinase inhibitors (imatinib) was a turning point in the treatment of chronic myeloid leukemia (CML). However, some patients are either refractory to imatinib or may develop relapse. Such resistance is often associated with mutations in tyrosine kinase domain of the chimeric BCR::ABL1 gene which are characteristic of CML. Moreover, approximately 100 point mutations have been identified in CML patients resistant to imatinib therapy, causing a single amino acid substitution in BCR-ABL1 gene. Some of these mutations affect the binding sites of imatinib with the chimeric tyrosine kinase BCR-ABL1. Other mutations affect the regulatory regions of the kinase domain, resulting in reduced sensitivity to imatinib and aberrant kinase activity. Early detection of BCR::ABL1 mutations allows choosing a second line of therapy. In particular, the patients with T315I mutation are resistant to all currently used tyrosine kinase inhibitors, and allogeneic hematopoietic stem cell transplantation is the treatment of choice.

Our set of reagents allows detection of T315I mutation in tyrosine kinase domain of the BCR-ABL1 chimeric gene in clinical material of patients with chronic myeloid leukemia by polymerase chain reaction (PCR) with real-time hybridization-fluorescence detection, as well as monitoring the efficiency of therapy by assessing minimal residual disease (MRD) scores.
