ChimeriX is a system for determining donor chimerism after allogeneic hematopoietic stem cell transplantation (allo-HSCT) based on pattern analysis of highly polymorphic markers in donor-recipient pairs. The appropriate differences are monitored over the post-transplant period with product separation by capillary gel electrophoresis. ChimeriX genetic markers are distributed on 12 chromosomes and represent highly polymorphic microsatellite repeats (STRs). This approach provides a high probability of identifying informative markers by DNA fragment analysis in the donor-recipient pairs. The ChimeriX-3 system employs 15 markers: D8S639, HUMVWA31, D11S488, D19S246, D4S2366, D12S1046, D13S117, D16S539. The test system also covers polymorphisms of the amelogenin gene, an informative marker for the donor and recipient who differ in gender. The primers are labeled with 6-FAM, R6G or ROX fluorescent dyes.

The minimal concentration of genomic DNA for analysis is 200 pg, with optimal range of 0.2-4.0 ng of DNA per reaction under standard conditions.

Polymerase chain reaction (PCR) is carried out on solid-state thermocyclers of any manufacturer, however, the reaction conditions below are adapted for Bio-Rad, ThermoScientific (Life Technologies), Biometra cyclers. For capillary gel electrophoresis, Life Technologies genetic analyzers (3130, 3500, 3500xl) are recommended, using POP7 polymer.

For the fragment analysis, GeneMapper, Peak Scanner, or GeneMarker software is recommended.
